Glasdegib Maleate

Introduction 

Glasdegib, sold under the name Daurismo, maybe a medication for the treatment of newly diagnosed acute myeloid leukaemia (AML) in adults older than 75 years or those that have comorbidities that preclude the utilization of intensive induction chemotherapy. One can take it orally and employed together with low-dose cytarabine. In fact, it is a small molecule inhibitor of sonic hedgehog, which is a protein overexpressed in many types of cancer. It also inhibits the sonic hedgehog receptor smoothened (SMO), as do most drugs in its class.

Glasdegib is in fact, in a class of medications called hedgehog pathway inhibitors. It however works by blocking the action of a protein that signals cancer cells to multiply. This helps stop or slow the spread of cancer cells. The recommended dose of glasdegib is 100 mg orally once daily on days 1 to twenty-eight together with cytarabine 20 mg subcutaneously twice daily on days 1 to 10 of every 28-day cycle within the absence of unacceptable toxicity or loss of disease control.

US approved Glasdegib for medical use in December 2018. The US Food and Drug Administration (FDA) also granted it priority review and orphan drug designation. It was, in fact, granted orphan drug designation by the European Medicines Agency (EMA) in October 2017. Also, Glasdegib was approved for medical use in the European Union in June 2020.

Glasdegib comes as a tablet to require orally. It is usually given with or without food once each day for a minimum of 6 months, or as long as your doctor recommends treatment. Take glasdegib at around the same time a day. Follow the directions on your prescription label carefully, and ask your doctor or pharmacist to elucidate any part you are doing not understand. However, take glasdegib exactly as directed. Do not take more or less of it or take it more often than prescribed by your doctor. Swallow the tablets whole; don’t split, chew, or crush them. If you vomit after taking glasdegib, don’t take another dose. Furthermore, continue your regular dosing schedule.

Your doctor may have to interrupt your treatment, reduce your dose, or stop your treatment counting on your response to the medication and any side effects that you simply experience. Be sure to inform your doctor how you’re feeling during your treatment with glasdegib. Take the missed dose as soon as you commit it to memory. However, if it is less than 12 hours until your next dose, skip the missed dose and continue your regular dosing schedule. Do not take a double dose to form up for a missed one.

Side effects:

  • pain or sores in your mouth or throat
  • decreased appetite
  • change in the way things taste
  • weight loss
  • headache
  • rash
  • swelling of hands or legs
  • hair loss
  • toothache

Some side effects can be serious. If you experience any of those symptoms, call your doctor immediately:

  • feeling faint, lightheaded, or dizzy; fast or irregular heartbeat
  • unusual bruising or bleeding
  • fever alone or along with chills, weakness. or other signs of infection
  • decreased urination
  • chest pain
  • Symptoms of overdose may include:
  • nausea
  • vomiting
  • extreme tiredness
  • dizziness
